Job description

Adoptive therapy with T cells has shown clinical successes in advanced tumors. For these trials to become future proof, particularly in the field of solid tumors, there is a need for rational strategies to further enhance safety and anti-tumor efficacy. At Erasmus MC, we are currently performing a first-in-man phase I/II trial to demonstrate safety and efficacy of T-cells that are gene-engineered with an anti-MAGE-C2 (MC2) T-cell receptor (TCR) in advanced melanoma.

The T-cell-based ATMP (Advanced Medicinal Therapeutic Product) is administered to patients following pre-treatment with epigenetic drugs (no lympho-ablative chemotherapy) and is supported by low dose interleukin-2. Notably, the TCR T-cells are generated according to new Good Manufacturing Practices (GMP) protocol, and harbor a young phenotype. For more information, see this link.

Erasmus MC is finalizing an accelerated titration scheme (phase I) and anticipates to continue with a recommended phase 2 dose, to which end, researchers are scheduling a trans-national trial together with Charite, Berlin, and AP-HP, Paris, as part of an European initiative (EUCCAT: European Consortium for Cellular Cancer Therapies within the European University Hospital Alliance (EUHA)).

During this clinical trial, the candidate researcher will coordinate the production of the T-cell ATMP, and perform research into the introduction of new, larger scale manufacturing methodologies for T-cell-based ATMPs. The candidate will team up with experienced technicians, researchers and clinical oncologists at the department of Medical Oncology, as well as specialists at the ATMP facility of Erasmus MC. In addition, the candidate will write quality reports and regulatory documents as well as disseminate findings during meetings and conferences.
